"Work" in Australia?

So what, exactly, are we doing in Australia?  The answer is a little more complex than a deep desire to understand cricket or a scheme to get rich by sneaking gallons of eucalyptus oil back into the US.



My sabbatical work is focused on a biological process known as RNA interference. This phenomenon is now known to occur in microorganisms, plants, animals, and humans, but was only discovered in the late 1990’s through studies with worms. The principle scientists behind this discovery were awarded a Nobel Prize in 2006, and the interest in using RNA interference to combat human disease continues to grow today.

So what is it? RNA interference is a mechanism that allows for gene “silencing”. Normally, messenger RNA is used to translate genetic information encoded in DNA to proteins that actually do the work of the cell. Gene silencing occurs when complementary RNA binds to messenger RNA and causes it to be degraded, preventing the protein from being produced. 



If you are interested and have 4 minutes, this video from NOVA does an excellent job of explaining the biological context and the mechanics of RNAi.

My interest in RNAi stems from working to solve the tricky problem of metabolic disorders in early lactation dairy cows. Despite tons of research on this problem, we still have an estimated 40-50% of dairy cows dealing with at least mild forms of these disorders, which include fatty liver and ketosis. In recent years, many groups (including ours) have published studies documenting changes in gene expression in the liver that correspond with metabolic problems. However, without having the tools to directly alter these processes, there is no way to determine whether these changes are actually contributing to the problem or if they are part of a rescue/survival strategy.  Without more direct evidence about the signals driving the disease process in these animals, our attempts to find a solution are based mostly on guesswork.



In the long-term, my goal is to develop a product that would use RNA interference to alter liver function in dairy cows at the start of lactation to prevent metabolic disorders. Before that can even be attempted, though, a lot of background work is needed. For the coming 5+ months I will be working with an outstanding team of scientists in CSIRO, Australia’s government research agency. This team is based at the high-security Australian Animal Health Laboratory in Geelong, in part because they are working on the use of RNA interference to combat some very serious viruses such as Ebola. My work during this sabbatical will be focused on evaluating different nanoparticles that can help carry small RNA molecules into target cells in the body, a critical and tricky aspect of using RNAi in animals (as opposed to cultured cells). Much to Sarah’s relief, I will not be working with any interesting pathogens. :)

Day-to-day, right now I am brushing up on some lab skills that have not been used much for the last 5 years, while also picking up lots of minutia that is critical to successfully using new techniques, but is difficult to learn by reading papers.   Hopefully this will lead to good things down the road!

I do need to acknowledge K-State for letting me have this sabbatical and the Fulbright program for helping us to pay the bills so we can be here.  This is an amazing opportunity and we are very thankful for it.

Mystery tree and 2 Australian animal stories by Kiernan

We found this tree in the backyard of the house we’re staying in. I think it’s a eucalyptus.  (Type of eucalyptus) blackbutt.  There are 700 types of eucalyptus trees!  Do you want to know more information?  Yes?  Okay.  Then here we go!  The leaves are light green.  The bark is kind of lightish brown.
The leaves are toxic, but koalas are tolerant.  Blackbutts can be used to soak up swamps.🎑😁Kiernan






















The Platypus Who Saved Their Eggs

It was a stormy day. Two platypus climbed up in their home. The eggs were gone! Suddenly, the eggs were in sight. They were floating away! The platypus made a dash for it (actually, a swim for it). Finally, they caught up to the eggs. They grabbed them in their bills and brought them back to their home. The End.

Once upon a time Black Kangaroo was hopping around and his archenemy (the dingo) spotted him. He thought, "I'll trip him in the mud with this stick." Now Kangaroo came along and tripped on the stick into the mud. Dingo had been hiding in the bush and he came out. Kangaroo said, "I should have known dingo boy thinks he's so smart." "Well, I am." says Dingo. "Oh yeah, watch this." said Kangaroo. With a mighty pull he popped out and hopped away as fast as he could. But he was brown. The End.

Lorne, Great Ocean Road and Our Home until July

Phew! Seems like we've been busy with schoolwork, moving around to different rentals, traveling and exploring on the weekend and FINALLY moving into our permanent place while here.

We went to Lorne last weekend. It's about 65 km from Geelong and is a town on the Great Ocean Road. This was further than Torquay/Bells Beach, so the road got much more interesting. It's a very windy road and narrow with a dropoff to the ocean in some places. It is gorgeous!! We took turns driving, so we could both stare. We ate lunch at a place in Lorne that overlooked the local beach. It was rather busy, but too chilly for the Kansans! After lunch we drove to the entrance to the trail that lead to Erskine Falls. Erskine Falls is in the Great Otway National Park (called The Otways by locals) and is one of the most popular falls to visit because its easy to access. It was a short walk downhill via stairs to see this gorgeous view!

On the drive back we stopped along one of the beaches and looked through some rock pools. We saw a few starfish, snails and lots of shells. I also caught a glimpse of one fish (looked like a blenny), but it swam out of view once it saw me.

Galah
Barry and the kids have done a bit of geocaching. There are many to find in this area! When we were at the Newcomb rental last week we saw parrots up in one of the trees. That was a pretty fun experience to notice them and then figure out what they were. They are called galahs. Not the most colorful bird you find in this area, but still very interesting and different to see in the wild.

On Tuesday, January 20th we moved into our permanent place in Ocean Grove, Victoria. If you need our mailing address please let me know and I'll send it to you. It's a 4 bedroom, 2 bathroom home with a nice fenced in backyard. There's a veggie patch, trees and a large grill. Bring on the barbie!! The kids each get their own bedroom, so they are happy about that. There's a washer and dryer (dryers aren't as common here). Most people hang their laundry to dry. There's a nice big clothesline outside, so I've been hanging most laundry and using the dryer if I run out of daylight. The home came furnished, so other than some bedding and towel purchases we've had everything we need. It's nice to have some room to spread out.

On Wednesday we went back to Ocean Grove Beach to enjoy the waves. It was low tide at the time, so the kids were kind of amazed at how far out the water was. Hannah and Kiernan tried out the boogie boards we found in the shed here. They both had some practice at Cherry Grove Beach in South Carolina a year and a half ago, so they caught on quick here and can ride the waves pretty well. We also had fun building a sand castle village.

 Thursday evening we had a spider incident! I noticed mid-afternoon what appeared to be a very large dead spider between the screen and glass in one of windows. As soon as Barry was home we showed him. After dinner, we decided it was best to at least get it out of the window. I wasn't certain it was dead so I wiggled the screen some and it moved. I about LOST IT! The screen is broken at the bottom, so if it wanted it could have come inside-ummm, yikes!! Barry decided to take a broom outside and get it out that way, but it was so scrunched up at the top of the window that we had to spray it a bit through the screen to get it to drop out of the window. Then it kind of crawled off and it seemed to watch us for a bit. ICKY! It was a Huntsman spider....look them up if you want to learn more. They are pretty much harmless to people, other than causing a serious scare because of their size. They are just so darn huge. Apparently they like dark corners and hiding places, so it is common that they are found in cars too. A common location is above the visor, so when you put it down they fall on you. So, now I'm busy checking the visors before I get in the car too. I also examine the windows in our house a lot....I really don't want any unwanted visitors IN the house! Sadly, I doubt this is our last spider story. This place is teeming with critters. On that note, I've not seen a snake yet.

On Friday, Barry texted me on the way to work to let me know that our car had overheated and was on the side of the road. Sure hoping we didn't buy a lemon! After a $120 tow it looks to just need a new radiator (or something like that). The local mechanic had just serviced the car 2 days before, so we already know him. He gave us a loaner car for the weekend (it's tiny, but we all fit). Just part of the adventure, right?!

We've been settling in to the new place well. We keep the kids busy with a few chores around here and sneak in as many sunsets as we can. Barry takes the car to get to work in Geelong (which is a 20 minute commute for him). The kids and I have about a 10-15 minute walk to the business district of Ocean Grove, so we've already walked to the library, Target Country, and a toy store. The walk is easy and makes for a fun adventure. We finally got a library membership and have taken full advantage of it already! Hannah and I even went back today to return some and checkout more.

Exploring Geelong region

We started off our weekend at a little cafe in Geelong that Barry discovered when he was here in September. He said he thought I would love it and oh, was he right!! From the front of the cafe it looks like any other little coffee shop with a small seating area inside. Once inside though you walk out through the back of the cafe and it's a little hidden oasis! Gorgeous plants and lots of bright sunshine through the covered roof. Colorful seating, leather furniture and pretty knick-knacks everywhere. I could have sat there for hours. I tried to take a few pictures to show everyone (without seeming creepy). Took the top one over my shoulder.

We shared a nice selection of pastries and specialty drinks. They hand out stuffed animals instead of numbers when you order (so they can bring your order to you). Lydia thought this was awesome! Coffee is a bit different here. You can't often find drip coffee, so you have to order a long black or a latte. According to Wikipedia, "a long black is a style of coffee, most commonly found in Australia, New Zealand and Brazil. It is made by pouring a double-shot of espresso over hot water." It's similar to an Americano for all you coffee aficionados out there! What about coffee pots in hotels and homes? No coffee pots in hotels or the family home we are staying in now. Every place has had a kettle for hot water and instant coffee and tea seems to be the usual. We may splurge and purchase a coffee pot in our permanent rental that we move into next week (if they don't have one). Must have our coffee!!

After filling up on drinks and goodies we took the kids to Kids Runamuk, which is an indoor playground. They loved it! There was a toddler area, but Lydia wasn't having that. She wanted to play in the big kid area. She loved peeking at us through the window up in the playground. Funny girl.

After the kids were worn out we headed towards the coastal village of Torquay and the famous Bells Beach. Torquay is also the start of the Great Ocean Road and is known as one of the world's great scenic drives. We plan to do the full drive at a later time. Bells Beach is a famous surfing beach and is not for swimming by Kansans! A natural underwater rock platform contributes to the excellent surfing conditions at Bells. There is an international surfing competition held at Bells at Easter, which apparently brings thousands of tourists (not sure if we will go or not). The waves were huge on this beach! We enjoyed watching the surfers catch the waves. There was definitely some talent out there!

Queenscliff was the next stop on our journey. It's another coastal village in the region. This village was beautiful and my favorite so far. Tons of quaint little shops and gives a European vibe. We ate a late lunch at Rolling Pin Cakes and Pies, which had a banner that said it had the 2013/14 Australia's Best Pie. Meat pies are common in Australia and similar to chicken pot pie, but many more flavors to choose from. We've discovered that Hannah loves the flavor of lamb, so we tend to order something with lamb everywhere we go. Lamb is much more common in Australia than in the USA.

The kids were pretty tired after that (ok, so were the parents), so we headed back to our hotel for a bit. We took an evening walk along the bay and enjoyed seeing and hearing the sights and sounds of the bay. These black swans were fun to watch.

On Sunday we went to Mass at St. Mary of the Angels Basilica in Geelong. We've done some communicating with the priest at this church, so after Mass we had the opportunity to connect with him in person. He was very welcoming and offered to help in any way we needed. His name is Father Kevin Dillon, and he said he's been to the U.S. 7 (I think) times. Several of those trips were to Hawaii as they do service projects there, but he'd also been to San Franscisco and New York. Never Kansas though.

After Mass we went to the waterfront to catch a light lunch and then took the kids on their first ride on the Geelong Carousel.

We checked into our Airbnb rental for the week. It's a family home (the family is away on holiday), so it comes fully furnished. The family has 2 young boys, so there are plenty of toys around. It's got a big backyard, with a playground and sand pit. Kids are thrilled about that!! There's also a large covered patio with table and chairs, so we've been having most of our meals outdoors. We enjoyed going to the grocery store and stocking up for a week! I've been making some of our favorite meals, as we were ready for a break from eating out.

Hannah has declared homeschool so much fun! She misses her friends, but in a happy moment said that she enjoys hanging out with mom all the time. I'll take it!


Finally, I know you were all wanting to see our sweet new ride. I wanted Barry to get this car for us, so we could drive the coastline in style. Isn't it pretty?


Unfortunately, Mr. Practical thought we better get this Holden Commodore Berlina instead (something about luggage fitting and children's safety). All joking aside, it's been a decent car so far and the luggage does fit (well, minus the 3 bags we stashed at a friend's place). 


It's a rainy Tuesday here, so we are stuck inside. Apparently this much rain is highly unusual. There are huge puddles everywhere and it is the first time we've seen that. Hoping the sun returns soon.

Food and good fun!

Wednesday evening we tried out the Little Creatures Brewery here in Geelong. Barry had already been to this place when he was here in Australia in September 2014. It ranks high as a place to take the kids for dinner. I know, weird....since it's a brewery! It was a rainy evening and we knew they had an outdoor space for kids, but once we arrived we knew we'd be better off inside (it did start pouring rain not long after we sat inside). It was great. The inside seating still feels like outside. It's built in an old carpet factory, so very unique feel. There were lots of great seats and umbrellas outside and a giant sandbox for kids, so we'll have to go back when the weather is better.

Barry and I always share when we are traveling so that we can try out many items on the menu. The grilled corn with chipotle butter sure reminded me of Taco Lucha in Manhattan, KS. Oh, how we will miss that place for the next six months!

We've spent a fair bit of time finding parks to stretch our legs in between schoolwork and Frozen (that's what Lydia likes to do with her spare time-whether it's watching the movie or pretending to be Elsa). The sea air has brought out all the curls in Miss Lydia's hair. It's pretty wild and nothing tames it! So cute!

Last night (Thursday) it was pretty rainy, but good enough to go exploring around the city again. We stopped in a toy store to pick up a few new things to keep us busy. Then we went to Eastern Beach in Geelong. There is a great playground! I captured a few pictures of the city while there. There's a children's pool, beach and playground all right there, so when the weather is warmer we'll be heading back. There were a few brave souls swimming in the pool with their kids, but I noticed they all had wetsuits on.

We are looking forward to the weekend as Barry will be with us more! We'll blog after our weekend and tell you what we discovered. I've also asked Barry to blog soon explaining what he is up to on his sabbatical, just in case you are wondering. Plus, I'm sure this blog will get quite boring if I do all the writing. We will be moving into a new place on Sunday and will be there for a little over a week. Then on Tuesday, Jan. 20th we finally move into our permanent home through July.

House, hotels, homeschool and Holden Commodore

Hello again! We found out yesterday that we've been approved/selected (whatever it is) for the rental house in Ocean Grove, Victoria. We are so excited! It's a fully furnished 4 bedroom, 2 bathroom place with fenced in yard, garage, beautiful trees, etc. We have plenty of space for visitors and each kid will have their own room when we don't have visitors. It's in what is considered "old" Ocean Grove and the street is quiet. We met the owners when we viewed it and they said there are many kids on the street. Start booking your tickets to come visit now! Plus, Ocean Grove Beach is about a mile from the house. Walking distance if we want!

We drove in to Ocean Grove to sign paperwork for the house yesterday. While there we hit up the local ice cream shop for a quick treat. Ocean Grove is a pretty small community, but it has many neat shops and feels very clean and friendly. There is a really nice looking library just a few blocks from the house, so I suspect the kids and I will be stopping in frequently. It is about a 25 minute commute for Barry to get to work, but we figured being close to the beach was worth the trade-off of driving!


We can't move into the house until Tuesday, January 20th, so unfortunately we have to live out of our suitcases for a couple of weeks. We are staying at a nice 2-bedroom apartment (lacking a full kitchen) on the bay in Geelong for this week. We've got a home rented next week on an acre and boasts 40 fruit trees. In the meantime, we are busy having picnics on the hotel room floor for some meals. The kids think this is such fun!

We started school yesterday as well. The kids are getting used to pretending Mom's a teacher, and we are getting in a groove to accomplish schoolwork each day. It's hard with the nice weather to take it seriously, but we had a better day today than yesterday. We managed to get most work done this morning, including taking a break to make this tower out of Connectagons.

We also took a break to walk along the bay and stop at a park to play for a while. Gorgeous weather again today-80 degrees or so. Supposed to be a HOT one again tomorrow, then the rain is coming.

The most exciting and terrifying part of the day was picking up our "new" 2002 Holden Commodore station wagon. I haven't been driving the rental car, as we would have had to pay extra to have 2 drivers. Therefore, my first experience of driving in Australia (on the left side of the road and with the wheel on the other side of the car) was when I drove our car off the lot. We had to drive the rental car back into the Melbourne airport area to drop it off, so I followed Barry in our new beast. All went well and we made it back to Geelong in one piece. We'll post some other pics of the car once we snap them.

We rounded out the night by stopping for pizza and then the older two kids went swimming in the hotel pool. Feeling more settled in all the time. We are looking forward to moving into the house in Ocean Grove for many reasons, but we'll spend the next two weeks enjoying our rentals as well.
